How to Take Axinix (Axitinib)
Route: Oral, taken by mouth
Standard dose: 5mg taken orally twice daily (approximately 12 hours apart). Treatment is continued until disease progression or unacceptable toxicity.
Dose escalation: For patients who tolerate the starting dose with no adverse reactions above Grade 2 severity, whose blood pressure is not above 150/90 mmHg, and who are not receiving antihypertensive medication, the dose may be increased stepwise to 7mg twice daily and then to a maximum of 10mg twice daily.
Dose reduction: If side effects require it, the dose may be reduced stepwise to 3mg twice daily and then to 2mg twice daily. The 1mg tablet is used in combination with the 5mg tablet to achieve the exact required dose at each step.
With food: Can be taken with or without food. The tablets should be swallowed whole with a glass of water and must not be crushed or split.
If a dose is missed: If a dose is missed or vomiting occurs after taking a dose, an additional dose should not be taken. The next prescribed dose should be taken at the usual scheduled time.
CYP3A4 and CYP3A5 interactions: The dose should be reduced by approximately half (for example, from 5mg twice daily to 2mg to 3mg twice daily) when co-administering with strong CYP3A4 or CYP3A5 inhibitors such as ketoconazole.
